چکیده

The theory of antibody-dependent enhancement plays an important role in the dengue virus infection. However, its molecular mechanism is not clearly studied partially due to lack of a sensitive assay to determine the dengue virus-infected cells. We developed a flow cytometric assay with anti-dengue antibody intracellular staining on dengue virus-infected cells. Both anti-E and anti-prM Abs could enhance the dengue virus infection. The anti-prM Ab not only enhanced the dengue virus infected cell mass, but also increased the dengue virus protein synthesis within the cells. The effect of anti-prM Abmediated enhancement on dengue virus infection is serotype-independent. We concluded that the target cell-based flow cytometry with anti-dengue antibody intracellular staining on dengue virusinfected cells is a sensitive assay to detect the dengue virus infected cells and to evaluate the effect of enhancing antibody on dengue virus infection on cell lines or human primary monocytes.

Source and Purity of Dengue-Viral Preparations Impact Requirement for Enhancing Antibody to Induce Elevated IL-1β Secretion: A Primary Human Monocyte Model

Dengue virus is a major global health threat and can lead to life-threatening hemorrhagic complications due to immune activation and cytokine production. Cross-reactive antibodies to an earlier dengue virus infection are a recognized risk factor for severe disease. These antibodies bind heterologous dengue serotypes and enhance infection into Fc-receptor-bearing cells, a process known as antibo...

The cytokine response of U937-derived macrophages infected through antibody-dependent enhancement of dengue virus disrupts cell apical-junction complexes and increases vascular permeability.

Severe dengue (SD) is a life-threatening complication of dengue that includes vascular permeability syndrome (VPS) and respiratory distress. Secondary infections are considered a risk factor for developing SD, presumably through a mechanism called antibody-dependent enhancement (ADE). Despite extensive studies, the molecular bases of how ADE contributes to SD and VPS are largely unknown. This w...

Flow cytometric analysis of dengue virus-infected cells in peripheral blood.

With the development of permeabilization techniques in flow cytometry and the availability of various monoclonal antibodies (MAbs) that specifically bind with cell surface and intracellular antigens, it is now possible to use flow cytometric assay to identify dengue virus (DEN) infected cells in peripheral blood. Blood samples were analyzed using phycoerythrin (PE) labeled anti-CD3, anti-CD14, ...

Dramatic caspase-dependent apoptosis in antibody-enhanced dengue virus infection of human mast cells.

Severe forms of dengue virus disease, known as dengue hemorrhagic fever and dengue shock syndrome, result from an aberrant immune response involving antibody-dependent enhancement of infection, thrombocytopenia, and a loss of vascular integrity, culminating in hemorrhage, shock, and in some cases, death. Several studies have indicated that dengue virus infection results in the induction of apop...

Primary human splenic macrophages, but not T or B cells, are the principal target cells for dengue virus infection in vitro.

Understanding the pathogenesis of dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) requires the precise identification of dengue virus (DV)-permissive target cells. In a previous study using unfractionated human peripheral blood mononuclear cells, we found that monocytes, but not B or T cells, were the principal DV-permissive cells in the absence of DV-immune pooled human sera (PH...
